After a fresh install on a Mac Big Sur, (Current channel (preview)) I get a dialog when opening Excel or Word. First "What's new in Excel", then the dialog "Your privacy option". I can click on the OK-button, but nothing happens. When closing the window/dialog then also Excel is closed.

I had the same problem running Big Sur. In my case, I have just changed licensing company after changing jobs, so that may be the reason for the problems. I followed @TobiasHammarstrand's suggestion of using the "License removal tool" and then reactivate Microsoft 365, and voila - it worked!

    This worked for me.

The OK button does not work in my case (simply not clickable) and the application crashes when i close the window (x).

The "OK" button on the Privacy Option message screens work for you? I click OK and nothing happens... If I close the window (red X) the message screen goes away and the application starts. But - it repeats each time I launch the application.
