My PC is working really normal before update. After I update my windows 10 the system show this message.
C:\WINDOWS\system32\config\systemprofile\Desktop is unavailable. If the location is on this PC, make sure the device or drive is connected or the disc is inserted, and then try again. If the location is on a network, make sure you're connected to the network or internet, and then try again. If the location still can't be found, it might have been moved or deleted. - OK
Then I click ok after that I only see recycle bin and task bar, which can not do anything background is black, so I restart by using ctrl+alt+delete and run safe mode(safe mode with network).
In safe mode it has no This PC it just has recycle bin and some program and I can't open any file location of any program after that I turn off my pc for 30 sec then open it the result is
C:\WINDOWS\system32\config\systemprofile\Desktop is not accessible
Access is denied. -OK
Only recycle bin and task bar and black background again.

Open Windows File Explorer
On the View Ribbon, check the box marked 'Hidden Items'
C:\users\Default\
Right click and copy the Desktop folder contained in the Default Folder
Navigate to: C:\Windows\system32\config\systemprofile
Right click and Paste the Desktop folder into the systemprofile folder
Restart (not shut sown) your PC
Okay, this looks like it is working. It will remove all files installed that didn't come with the computer but keep my personal files (so it says). It looks like I won't be reinstalling McAfee, as it appears the virus detection programs are to blame in most cases. Looks to me like Microsoft would have windows check for a third party program and say it needs to be disabled.

I had/have this same problem so I took out the C: drive and plugged into another computer. I navigated to C:\WINDOWS\system32\config\systemprofile\Desktop and discovered that the systemprofile folder didn't exist. I did see the desktopfolder, but it was in the config folder, so a level up from where it should be. I figured I'd make a systemprofile folder and drop the desktop folder into it to see if that worked. However, when I went to try it, I got this crazy message:
As you can see, I was attempting to rename new folder to systemprofile, but it was giving me a warning that there was already a folder named desktop, even though I didn't type desktop! This made me think that somehow it was labelled incorrectly internally. Sure enough the missing folders are indeed within the desktop folder (including another desktop folder). Instead of making a new desktop folder, I renamed the parent one to systemprofile. I haven't plugged the drive back into it's machine yet, but I am hoping this will fix the issue.

For anyone not helped by the "solution" marked above, this is the next step.
Basically this is a failed Windows Update install. The solution I found was to reinstall the newest Windows patch over the top, and it fixed everything and I was back to before the issue happened.

2.On a working system, format a thumb drive to NTFS (file size restriction) and copy the contents of the .ISO into a folder.
3.On defective computer, hit CTRL+SHIFT+ESC to bring up task manager. You may have to click "more options" to see menu.
4.Under File menu, top left, click "run new task", and "browse". This is the only way I was able to access my file system.
5.Find your thumb drive with extracted Windows patch, and run the Setup file, clicking the box to "run as Administrator". This will run the update again.
After reboot you will be good to go. I've seen this work for two systems, and about 6 here on the forums.
