Usually, running the OneCare scanner restores the system's ability to update.
It will provide a list of what it detected, and attempt to remove it.
IF it detected numerous malwares, then the system has been serverly compromised and a format/reinstall may be in order.
The Scanner for Vista is different than the one for XP, Windows 2000, etc.

Specifically, which malwares were detected when MBAM, SAS, WinPatrol, and Avast were run ?
I know of no conflicts between iTunes software and Windows Update.
The 0x80072EFD error code means that the system can not connect to the update servers and is usually related to an issue with the Windows Update Agent ... either it is not installed properly or a related .dll is not registered. Malware can be the cause of both,
but so can system inconsistencies. Which is why I asked you if the System Update Readiness Tool [CheckSUR] was used.
Has Service Pack 2 been applied ? If so, then the message you received when attempting to run CheckSUR is a
known bug .
Does the system have any connectivity issues or is it only related to Windows Update ?
